It possesses DH/PH domains responsible for its GEF activity and SH3, coiled-coil, and calponin homology domains for scaffolding and protein-protein interactions[1][2][3]. ARHGEF6 regulates actin cytoskeleton remodeling, cell adhesion, and motility, and is essential for normal cognitive development and hair cell stereocilia maintenance in the cochlea; mutations are linked to X-linked intellectual disability and sensorineural hearing loss[2][4]. It forms complexes with partners like p21-activated kinases (PAK1, PAK2, and PAK3) and GIT proteins, orchestrating local cellular signaling[1][3].
