Rho guanine nucleotide exchange factor 7 (ARHGEF7), also known as Beta-PIX (βPIX), is a multidomain cytoplasmic enzyme and signaling scaffold protein that activates Rho family GTPases, particularly Rac1 and Cdc42, by promoting GDP-GTP exchange[1][5]. It plays critical roles in the regulation of cell migration, cytoskeletal dynamics, focal adhesion maturation, and neuronal development, including dendrite branching, spine and synapse formation, and axon specification[1][3][5]. ARHGEF7 forms complexes with p21-activated kinases (PAKs), GIT1/2, and various synaptic adaptor proteins, coordinating local signal transduction and structural reorganization of cells. Experimental evidence indicates it is essential for early neuronal polarization and axon formation, acting upstream of small GTPases such as TC10 and Cdc42[3][1]. Its dysfunction or altered regulation is associated with neurological and immune disorders, and it remains a focus of research for its roles in cell migration, invasion, and disease[1][5].
