Rho-related BTB domain-containing protein 1 (RHOBTB1) is an atypical member of the Rho GTPase family distinguished by a unique multi-domain structure, comprising a GTPase domain, a proline-rich region, two BTB (broad complex, tramtrack, and bric-a-brac) domains, and a C-terminal domain[2][3][4]. Unlike canonical Rho GTPases that regulate the actin cytoskeleton and cell motility, RHOBTB1 functions as a tumor suppressor, with its loss or silencing associated with several epithelial cancers[2][4]. It is highly expressed in reproductive organs, muscle, and vascular tissues, and contributes to signal transduction, the organization of actin filaments, and, via its BTB domains, likely serves as an adaptor for ubiquitin ligase complexes[2][3][4]. RHOBTB1 also has emerging roles in vascular function and blood pressure regulation[4], and its expression is regulated by transcription factors (e.g., PPARγ) and microRNAs such as miR-31[2][3]. No direct drugs or clinical therapeutics are currently known to target RHOBTB1, and it is not considered a classical druggable therapeutic target (such as a receptor or enzyme), but it may serve as an important biomarker or molecular handle for researching cancer and cardiovascular disease biology[2][4].
