Overview

Rho-related GTP-binding protein RhoT2 (RHOT2/MIRO2) is a member of the RHO GTPase family, functioning as a mitochondrial outer membrane protein that regulates mitochondrial trafficking, distribution, and quality control within cells[2]. It acts as a molecular switch, cycling between active (GTP-bound) and inactive (GDP-bound) states, and participates in signal transduction pathways by interacting with a large network of regulatory proteins including guanine nucleotide exchange factors (GEFs), GTPase-activating proteins (GAPs), and downstream effectors[2]. Aberrations in RHOT2/MIRO2 signaling are implicated in cancer progression (especially cell migration and invasion) and in neurodegenerative diseases due to its essential role in mitochondrial dynamics[1][2]. Therapeutic targeting of RHO GTPases, including RHOT2, remains challenging due to the complexity and ubiquity of their signaling roles in normal physiology[1].
