Overview

Rho-related GTP-binding protein RhoV (RHOV) is an atypical member of the Rho family of small GTPases, sharing features with Rac and Cdc42 subfamilies. Distinct from classic Rho proteins, RhoV contains unique N- and C-terminal extensions and is post-translationally modified by palmitoylation but not prenylation. It regulates the dynamics of the actin cytoskeleton, particularly through participation in signal transduction pathways including the c-Jun N-terminal kinase (JNK) pathway. RhoV alters cell shape and cell adhesion, contributing to processes such as epithelial morphogenesis and heart development, but the detailed molecular mechanisms are still under investigation. No direct small molecule inhibitors or drugs targeting RhoV are currently known.
