Rhomboid domain-containing protein 3 (RHBDD3) is a membrane-associated protein predicted to possess serine-type endopeptidase activity and functions as a negative regulator of the activation of natural killer (NK) cells, particularly in the context of Toll-like receptor 3 (TLR3) stimulation. It is broadly expressed in immune cells and involved in feedback inhibition of inflammatory signaling pathways, including attenuation of MAPK signaling and regulation of protein degradation. RHBDD3 interacts with DNAX activation protein of 12 kDa (DAP12), promoting its proteasomal degradation and thereby modulating NK cell effector functions and contributing to immune homeostasis. Loss or dysfunction of RHBDD3 has been associated with increased NK cell activation and exacerbated inflammatory responses, especially in the liver, suggesting a potential role in protection against acute liver injury and possibly in the pathogenesis of liver cancers. As of now, no drugs are known to directly target RHBDD3, nor is it a standard biomarker or therapeutic target. However, its role in immune regulation and cancer biology highlights it as an emerging subject of interest in immunology and oncology research.
