Rhomboid-like protein 3 (RHBDL3) is a member of the rhomboid family of integral membrane proteins, characterized by its serine-type endopeptidase activity. This protein is involved in regulated intramembrane proteolysis—it cleaves substrates within or near their transmembrane domains inside the lipid bilayer, enabling the release of functional polypeptides from their membrane anchors. The rhomboid superfamily is broadly conserved across species and plays diverse roles in membrane processes, including protein quality control and potentially signaling pathways. RHBDL3 is predicted to reside within cellular membranes and is functionally related to other rhomboid proteases. Disease associations include benign mammary dysplasia, and gene essentiality data suggest it may have roles in cancer cell proliferation and survival.
