Overview

Rhomboid-related protein 2 (RHBDL2) is a human intramembrane serine protease of the rhomboid family, localized to the plasma membrane. It selectively cleaves specific type I transmembrane proteins—such as thrombomodulin, epidermal growth factor (EGF), interleukin-6 and -11 receptors, and ephrinB3—thereby releasing soluble fragments crucial for cell signaling. RHBDL2 plays roles in epithelial homeostasis, inflammatory signaling, and wound healing, and is implicated in pathology including cancer by modulating key signaling pathways (e.g., Notch, Wnt/β-catenin). Its enzymatic activity can be inhibited by certain chemical classes, such as sulfonamide compounds. Dysregulated RHBDL2 expression or function may contribute to tumor progression and altered immune responses.
