Rhophilin-associated tail protein 1-like (ROPN1L) is a member of the ropporin family, primarily expressed in sperm, where it functions as an essential component of the flagellar fibrous sheath. ROPN1L interacts with A-kinase anchoring protein 3 (AKAP3) via an amphipathic helix region and is important for maintaining sperm structural integrity and motility. The absence of ROPN1L (or its paralog ROPN1) disrupts fibrous sheath formation, impairs sperm motility, and leads to male infertility, seen in knockout mice. Unlike PKA, ROPN1L does not bind cAMP and appears to act as a regulatory subunit for AKAP scaffolds, partly compensating for each other with ROPN1. ROPN1L is also implicated in the motility of motile cilia in other cell types. Although disease associations have been noted for infertility and possible cancer susceptibility, ROPN1L is not a therapeutic target and there are no known drugs directly interacting with it[1][2][3].
