Overview

Rhox homeobox family member 2 (RHOXF2) is an X-linked transcription factor from the homeobox protein superfamily, preferentially expressed in human testis and implicated in male germ cell differentiation and spermatogenesis. RHOXF2 functions as a transcriptional repressor and is associated with the cell membrane and microtubules, localizing at the leading edge of migratory cells. It is classified as a cancer-testis antigen, exhibiting restricted expression in normal tissues other than testis but prominent expression in a wide range of tumor types, making it a promising target for cancer immunotherapy and a putative biomarker for cancer detection. Experimental models indicate RHOXF2 promotes cell proliferation and transformation, and its knockdown reduces growth in cancer cell lines such as gastric cancer. RHOXF2 may also regulate other members of the RHOX gene family, contributing to a cascade regulation in spermatogenesis. No drugs targeting RHOXF2 have been approved; research is ongoing for CT antigens in immunotherapeutic applications. Safety concerns center on the potential for autoimmune testicular damage and the need to avoid promoting oncogenesis.

Mechanism of action

No approved drugs, but immunotherapeutic approaches are plausible: T cell-redirecting cancer immunotherapies against cancer-testis antigens (including RHOXF2) are under exploration (as for other CT antigens like NY-ESO-1). Knockdown of RHOXF2 reduces growth in cancer cell lines.
