Overview

RHOXF1 antisense RNA 1 (RHOXF1-AS1) is a long non-coding RNA transcribed from the X chromosome, acting as an antisense transcript to the RHOXF1 homeobox gene[1][2][4][7]. It does not code for a protein but plays a regulatory role at the RNA level. RHOXF1-AS1 is expressed predominantly in certain tissues, including breast and thyroid, and at lower levels in placenta and ovary[6]. Overexpression experiments have shown that RHOXF1-AS1 affects the expression of genes involved in DNA replication, synthesis, and the cell cycle, promoting differentiation and reducing proliferation in human pluripotent stem cells[6]. Evidence suggests that it may act in the cytoplasm to mediate mRNA decay or inhibit translation, and might also serve as a scaffold in the nucleus to guide chromatin-modifying complexes for transcriptional regulation[6]. RHOXF1-AS1 has been linked to various cancers, particularly as a potential biomarker for prognosis in lung adenocarcinoma[8]. At present, it is not recognized as a classic drug target (receptor, enzyme, transporter, etc.), and there are no known drugs that specifically interact with RHOXF1-AS1.
