Ribonuclease A family member k6 (RNASE6) is a secreted, cationic antimicrobial enzyme in the RNase A superfamily, predominantly expressed by myeloid cells such as monocytes, macrophages, and neutrophils, and active in the urinary tract[1][2][3][4][6]. It exhibits broad-spectrum antimicrobial activity against both Gram-positive and Gram-negative bacteria via bacterial cell agglutination and membrane permeation, independent of its ribonuclease activity[1][2][3][7]. This protein contributes to host defense, particularly in maintaining urinary tract sterility, and may additionally play a role in neuroinflammatory processes associated with cognitive decline in select populations[1][3]. No drugs are currently known to specifically target RNASE6, and it does not presently act as a clinical pharmacological target[3][7].
