Overview

Ribonucleotide reductase (RR or RNR) is a crucial enzyme responsible for the de novo synthesis of deoxyribonucleotides from ribonucleotides. This reaction is essential for DNA synthesis and repair in all living organisms, making RR indispensable for cell proliferation and genome stability. Because RR is vital for cell proliferation, it is targeted by several anticancer drugs. Overexpression or dysregulation of certain RR subunits—especially RRM2—is associated with cancer progression and resistance mechanisms.
