Ribonucleotide reductase (RNR) and DNA polymerase are essential enzymes involved in DNA synthesis and repair. RNR catalyzes the rate-limiting step in the de novo synthesis of deoxyribonucleotides (dNTPs). DNA polymerase synthesizes new strands of DNA. They are critical targets for cytarabine, a chemotherapeutic agent. Cytarabine inhibits both enzymes, leading to impaired DNA synthesis and cell death, particularly affecting rapidly dividing leukemia cells.
Cytarabine is phosphorylated and incorporated into DNA, inhibiting DNA polymerases directly and ribonucleotide reductase indirectly, ultimately impairing DNA synthesis.
