Ribonucleotide reductase regulatory subunit M1 (RRM1) is the large, catalytic subunit of ribonucleotide reductase (RNR), an essential enzyme for DNA synthesis and repair. RNR catalyzes the conversion of ribonucleotides to deoxyribonucleotides, providing the building blocks required for DNA replication and repair in all dividing cells. It forms a hetero-oligomeric complex with smaller regulatory subunits—M2 or p53R2—to constitute active human ribonucleotide reductase. The M1/RRM1 subunit contains the catalytic site responsible for substrate reduction and two distinct allosteric sites that regulate both overall activity and substrate specificity through effector binding. Its expression and role in DNA synthesis make it a target in cancer therapy.
