Ribosomal biogenesis factor (RBIS) is a protein localized in the cytosol, nucleolus, and nucleoplasm that acts as a trans-acting factor crucial for the efficient assembly of the 40S ribosomal subunit. Its upregulation is correlated with poor prognosis and increased tumor aggressiveness in lung and prostate cancers, where it modulates ribosome biogenesis, cell proliferation, cell cycle progression, and apoptosis. Mechanistically, RBIS regulates rRNA synthesis and interacts with the ribosomal maturation factor GNL2, thereby influencing global protein synthesis and tumor cell sensitivity to chemotherapy. Its expression serves as a diagnostic and prognostic biomarker and presents opportunities for targeted cancer therapies, particularly in conjunction with drugs such as CX-5461 and gemcitabine.
