Overview

The Ribosomal peptidyl transferase center–nascent MYC polypeptide complex is a specialized state of the human ribosome during the translation of the MYC oncogene. This complex forms when the nascent MYC protein sequence interacts with the ribosomal exit tunnel, creating a unique structural pocket at the peptidyl transferase center (PTC). While MYC has long been considered undruggable due to its lack of traditional small-molecule binding sites, recent research has identified this specific ribosome-nascent chain complex as a viable therapeutic target. Small molecules can selectively bind to this complex, inducing translational stalling and preventing the production of the MYC protein. This approach offers a novel strategy for treating MYC-driven cancers by targeting the machinery of protein synthesis in a sequence-specific manner. By inhibiting the completion of MYC translation, these drugs effectively reduce the levels of one of the most potent drivers of tumor growth and survival.
