Overview

Ribosomal protein eL22-like 1 (RPL22L1) is a structural component of the 60S large ribosomal subunit, homologous to ribosomal protein L22. It is involved in ribosome assembly and helps maintain protein synthesis capacity, especially when its paralog RPL22 is deficient[1][2]. RPL22L1 is upregulated in various cancers, notably in lung adenocarcinoma, where high expression correlates with poor prognosis, increased proliferation, enhanced metastasis, and resistance to therapies. Mechanistically, RPL22L1 promotes tumor progression by suppressing the MDM2/p53 pathway and may facilitate other oncogenic pathways such as PI3K/Akt/mTOR and ERK signaling. Its expression is regulated by RPL22, which binds the RPL22L1 mRNA to repress its translation. RPL22L1 serves as both a functional marker for cancer progression and a potential therapeutic target, though no approved drugs currently target it directly[3][1][2].
