Overview

Ribosomal protein L13 (RPL13) mRNA encodes a structural component of the 60S large ribosomal subunit, which is essential for protein translation and ribosome biogenesis (UniProt P26373, NCBI Gene 6137). Beyond its fundamental role in the ribosome, the protein derived from RPL13 mRNA participates in the GAIT (Gamma Interferon-Activated Inhibitor of Translation) complex, which suppresses the translation of specific inflammatory genes (PubMed: 12606542). In clinical and laboratory research, RPL13 mRNA is frequently utilized as a stable housekeeping gene for normalizing gene expression data in quantitative PCR assays (PubMed: 20537170). However, its dysregulation has been linked to the progression of various malignancies, including gastric and breast carcinoma, where its overexpression may promote tumor cell proliferation and survival (PubMed: 33064245). While there are currently no FDA-approved drugs specifically targeting RPL13 mRNA, it is a subject of interest in experimental oncology using RNA interference-based approaches (PubMed: 31428242). The primary therapeutic challenge in targeting this molecule is its essentiality for basic cellular function across all healthy tissues, which presents a high risk for systemic toxicity (PubMed: 28655770).
