Ribosomal protein L24 (RPL24) is a structural component of the large (60S) ribosomal subunit, belonging to the L24E family of ribosomal proteins and located in the cytoplasm[1][2][4]. RPL24 is essential for protein synthesis by facilitating ribosome assembly and function. Beyond its classic ribosomal role, RPL24 modulates cap-dependent mRNA translation—overexpression in various cancers supports tumorigenesis via sustained protein production critical for cell survival and proliferation, while loss impairs 80S ribosome formation, reduces cell viability, and diminishes tumor cell growth[1]. RPL24 is also implicated in microRNA biogenesis through its interaction with components like DDX5, affecting the maturation of specific miR precursors[1]. Disease relevance includes a role in multiple cancers (breast, liver, laryngeal) and an association with immunodeficiency syndromes[2][3]. RPL24 has been explored experimentally as a potential therapeutic target, particularly for cancer, but there are no known direct small molecule or biologic drugs currently targeting it in clinical practice[1][3].
