Ribosomal protein L41 (RPL41) is a very small, basic component of the large ribosomal subunit, consisting of 25 amino acids[1][2][3]. It plays several vital roles in cellular biology, including maintenance of mitotic progression, proper chromosomal segregation, and cytoskeletal integrity by stabilizing polymerized tubulins[1][2]. Depletion of RPL41 disrupts spindle formation and centrosome integrity, leading to failed cytokinesis and aneuploidy—events linked to malignant transformation, supporting a tumor suppressor function for this protein[1][2]. RPL41 also promotes the proteasomal degradation of the stress-responsive transcription factor ATF4, causing cell cycle arrest and increased sensitivity of tumor cells to chemotherapy[1]. While often downregulated in cancer, RPL41 can also be upregulated in some malignancies, suggesting context-dependent regulation and a complex role in tumorigenesis[1][2][3].
