Overview

Ribosomal protein lateral stalk subunit P0 (RPLP0) is a component of the 60S subunit of the eukaryotic ribosome and is the functional equivalent of the E. coli L10 protein. It belongs to the L10P family of ribosomal proteins. The protein is a neutral phosphoprotein that interacts with P1 and P2 acidic phosphoproteins to form a pentameric complex (P0 as a monomer, P1 and P2 as dimers) as part of the ribosomal stalk, which is essential for protein synthesis. Located in the cytoplasm, P0 is critical in the elongation step of translation. Alternative splicing variants exist but encode the same protein. As with many ribosomal proteins, the gene has multiple pseudogenes dispersed throughout the genome. RPLP0 encodes a highly conserved ribosomal protein, structurally and functionally vital for the assembly and function of the ribosome. There is no evidence that RPLP0 is a direct target of therapeutic drugs or considered a canonical drug target such as a receptor, enzyme, or transporter. It is primarily studied for its fundamental role in ribosome biology and diseases associated with ribosomal dysfunction, not as a therapeutic target for drug intervention. No drugs are currently known to interact specifically with RPLP0, nor have mechanisms of action for drugs targeting this protein been described in the literature or drug databases. Furthermore, RPLP0 is not reported to be used as a clinical biomarker and has no specific safety/therapeutic concerns when modulated. Its primary associations with disease reflect its essential cellular roles.
