Overview

Ribosomal protein S15a pseudogene 12 (RPS15AP12) is a processed pseudogene transcribed as a long noncoding RNA that does not encode a functional protein but exerts regulatory effects in cancer. It acts as a molecular “sponge” for miR-96-3p, preventing this microRNA from repressing RPS15A, thus promoting RPS15A expression. RPS15AP12 enhances the growth, migration, and metastatic capacity of ovarian cancer cells and suppresses innate immune signaling through inhibition of RIG-I/MDA5 and downstream interferon pathways. RPS15AP12 is not a traditional therapeutic target as it is a noncoding pseudogene, but it may play a functional role in cancer progression and could potentially be considered for future biomarker or RNA-based therapeutic strategies.

02

Mechanism of action

Acts as a competitive endogenous RNA (ceRNA) sequestering miR-96-3p, thereby promoting the expression of its parent gene, RPS15A.

03

Biological functions

Regulation of gene expression via miRNA sponge activityPositive regulation of cell proliferation, migration, and invasion in ovarian cancer (acts as ceRNA for RPS15A)Modulation of innate immune response via negative regulation of RIG-I/MDA5 signaling
04

Disease associations

Cancer (specifically ovarian cancer, especially high-grade serous ovarian carcinoma)
05

Biomarkers

Overexpression of RPS15AP12 and its parent gene RPS15A in ovarian cancer may serve as a biomarker for tumor growth and metastasis potential, and for poor prognosis in ovarian cancer
