Ribosomal Protein S6 Kinase (S6K) is a family of serine/threonine kinases crucial for regulating protein synthesis, cell growth, and metabolism. The most studied forms, S6K1 and S6K2, are activated by the mTORC1 pathway in response to growth factors, nutrients, and energy status. S6Ks enhance translation initiation by phosphorylating rpS6 and influence glucose homeostasis and lipid synthesis. Dysregulation of S6K is linked to obesity, diabetes, aging, and cancer, making it a potential therapeutic target.
