Ribosomal protein S6 kinase 2 (S6K2) is a serine/threonine kinase that plays a critical role in cellular processes such as protein synthesis, cell growth, proliferation, and survival. It is activated downstream of growth factors and nutrients via signaling pathways like PI3K/Akt/mTOR and Ras/ERK-MAPK. S6K2 phosphorylates ribosomal protein S6 (rpS6) and eukaryotic translation initiation factor 4B (eIF4B), promoting increased global protein synthesis. Dysregulation of S6K2 has been implicated in several pathologies, including cancer, obesity, and diabetes, making it a potential therapeutic target. S6K2 directly interacts with the general transcription factor Yin Yang 1 (YY1), influencing transcriptional regulation through post-translational modification of YY1 itself. There is interest in developing selective inhibitors targeting deregulated signaling through this axis for therapeutic purposes.
