Overview

Ribosomal protein S6 kinase A1 is a member of the RSK family of serine/threonine kinases, encoded by the RPS6KA1 gene in humans. This kinase contains two nonidentical catalytic domains and phosphorylates various intracellular substrates, including components of the MAPK/ERK signaling pathway, ribosomal proteins, and key transcription factors such as CREB, ERα, and c-Fos. RPS6KA1 plays a central role in regulating cellular processes like growth, differentiation, proliferation, and survival. Its dysregulation has been linked to cancer and drug resistance, making it an important therapeutic target for small molecule kinase inhibitors and a biomarker for response/resistance in oncology and other pathologies.
