Ribosomal protein S7 is a structural component of the small (30S in prokaryotes, 40S in eukaryotes) ribosomal subunit, essential for the assembly and proper folding of the 16S rRNA (prokaryotes) or 18S rRNA (eukaryotes)[1][2][3][7][8]. Located near the decoding center of the ribosome, it initiates the assembly of the small subunit’s head domain and plays a crucial role in mRNA decoding and protein synthesis. S7 also binds its own mRNA, acting as a negative regulator of its translation (autogenous regulation), and mutations in RPS7 can disrupt ribosome biogenesis, leading to diseases such as Diamond–Blackfan anemia and, in mitochondrial forms, multi-organ pathologies[5][7][8]. Structurally, S7 is characterized by a bundle of six alpha-helices and a beta-hairpin, mediating intricate contacts with rRNA[1][2][4][6]. There is no evidence for direct interaction with drugs or use as a primary therapeutic target.
