Ribosomal RNA processing 36 (RRP36)

Target
RRP36
Molecular classification
RNA-binding protein, Preribosome assembly factor, Ribosome biogenesis factor
01

Overview

Ribosomal RNA processing 36 (RRP36) is a nucleolar protein required for early cleavages of 35S pre-rRNA, functioning as part of the 90S preribosome and interacting with pre-40S particles. Its role is conserved across eukaryotes, and loss of RRP36 leads to impaired 18S rRNA processing, reduced 40S ribosomal subunit production, nucleolar stress, and defects in cell growth. RRP36 is considered an essential factor for ribosome biogenesis but not a classical drug target.

Mechanism of action

Not applicable, as there are no drugs specifically targeting RRP36

03

Biological functions

Early processing of 35S pre-rRNAMaturation of 18S rRNAAssembly of the 40S ribosomal subunitNucleolar localization and interaction with 90S and pre-40S preribosomal particles
04

Disease associations

Defective ribosomal biogenesis can be linked to ribosomopathies, which are associated with cancer and disease phenotypesMutations or dysfunction in ribosome processing proteins can contribute to diseases related to cell proliferation and growth (such as cancer), though RRP36-specific effects are not well established
05

Safety considerations

Therapeutic modulation of core ribosome biogenesis factors such as RRP36 would likely result in broad cytotoxicity and loss of essential cellular function
06

Interacting drugs

None known or documented targeting RRP36 directly; drugs targeting ribosome biogenesis factors generally are not specific for RRP36
07

Biomarkers

None currently used for patient selection or efficacy monitoring
