Overview

Ribosomal RNA processing protein 1 homolog B (RRP1B) is a nucleolar protein that serves multiple roles in the cell, most notably in the processing of pre-ribosomal RNA during the formation of the large (60S) ribosomal subunit. RRP1B functions as a targeting and regulatory subunit for protein phosphatase 1 (PP1), directing PP1β and PP1γ isoforms to the nucleolus by specific and non-canonical interaction motifs[1][2][3][5]. Through its association with PP1, RRP1B participates in the reversible phosphorylation of factors involved in ribosome biogenesis and adapts ribosome production to cellular signals such as stress or changes in growth conditions[1][2][3]. RRP1B also acts as a transcriptional co-activator for E2F1, thereby positively regulating apoptosis in response to DNA damage[4]. In cancer biology, RRP1B has been reported as a modulator of metastasis and a possible biomarker for disease progression. There are currently no known approved drugs that directly target RRP1B, nor is RRP1B a recognized therapeutic target; its primary importance is in fundamental cell biology and as a potential biomarker[4].
