Overview

Ribosome-binding protein 1 (RRBP1) is a membrane protein predominantly located on the endoplasmic reticulum (ER) in eukaryotic cells[1][5]. It was initially described as a receptor mediating ribosome docking to the ER, though later evidence suggests that this activity is largely carried out by the Sec61 complex, while RRBP1 enhances the association of specific mRNAs with the ER independently of ribosomal activity[1]. RRBP1 plays a role in protein translation, translocation of nascent polypeptides, and the secretory pathway, likely contributing to ER structure and morphology. It has a recognized role in regulating the unfolded protein response and ER stress adaptation, making it particularly relevant in rapidly proliferating or secretory tumor cells[2][4]. RRBP1 is frequently overexpressed in certain cancers such as breast, lung, and colorectal, where it is associated with tumor progression and can serve as a prognostic biomarker[4]. It is also upregulated under stress conditions or upon exposure to chemotherapeutic agents such as paclitaxel and adriamycin in cancer cells, underscoring its importance in therapeutic resistance mechanisms[3]. Its potential as a therapeutic target is under investigation, especially in oncology[2][4].
