Overview

The ribosome biogenesis machinery refers to the complex network of molecular machines, including enzymes, ribonucleoproteins, chaperones, and regulatory factors, responsible for the stepwise synthesis, processing, assembly, and maturation of ribosomes in eukaryotic cells. Ribosome biogenesis begins in the nucleolus with RNA Polymerase I-driven transcription of precursor rRNAs, followed by processing, chemical modification, and assembly with ribosomal proteins transcribed by RNA Polymerase II. Small nucleolar RNAs (snoRNAs), multiple AAA-ATPases, GTPases, and other assembly factors mediate the maturation and correct assembly of the 40S and 60S ribosomal subunits, which are then exported to the cytoplasm for final assembly into functional 80S ribosomes. Proper ribosome biogenesis is crucial for translational capacity and cell growth, linking this machinery to key biological processes including development, proliferation, and apoptosis. Dysregulation of ribosome biogenesis machinery leads to diseases such as cancer and ribosomopathies, making it a promising but complex therapeutic target[1][3][4][5][6][7][9][10].
