Overview

Ribosome biogenesis protein NOP53 (commonly abbreviated as NOP53 and also known as GLTSCR2 or PICT-1) is a nucleolar factor essential for the biogenesis of the 60S ribosomal subunit in eukaryotic cells[1][2]. NOP53 plays a crucial role in integrating the 5S ribonucleoprotein particle into the large ribosomal subunit and participates in rRNA processing and maturation[1][2]. Beyond its direct role in ribosome assembly, NOP53 functions as a nucleolar stress sensor that regulates the tumor suppressor p53/TP53 pathway in response to ribosomal biogenesis perturbation, DNA damage, and cellular stress conditions[2]. It influences cell cycle arrest and apoptosis, partly by stabilizing the tumor suppressors p53/TP53 and PTEN, thereby antagonizing oncogenic PI3K-Akt/PKB signaling[2]. NOP53 has also been implicated as a tumor suppressor, with a role in regulating cell proliferation, apoptosis, and the nucleolar localization/function of proteins such as NPM1 and MYC[2]. No drugs or clinical agents directly targeting NOP53 have been reported.
