The Rift Valley fever virus (RVFV) envelope glycoprotein Gn is one of two major surface glycoproteins (Gn and Gc) encoded by the M segment of the RVFV genome. It is a type-I integral membrane protein that forms oligomers with Gc. Gn plays a critical role in recognizing and binding to host cell receptors, particularly DC-SIGN. Together with Gc, it mediates viral entry and membrane fusion. The cytoplasmic tail of Gn interacts with ribonucleoprotein complexes, driving efficient packaging of viral RNA segments. N-glycosylation at N438 enhances infectivity. Host factor UBR4 interacts with Gn and supports viral replication. Gn elicits strong antibody responses and is a target for neutralization.
