Overview

Rift Valley fever virus glycoprotein Gn is one of the two major structural envelope glycoproteins encoded by the M segment of the viral genome, the other being Gc[1][2][4]. Gn is embedded in the outer viral envelope and is exposed on the virion surface[2]. Together with Gc, Gn plays an essential role in attachment to and entry into host cells, with multiple lines of evidence supporting its direct involvement in host cell receptor binding and the assembly of viral particles[2][3][4]. The structure of the virion envelope is based on Gn-Gc heterodimers that form prominent surface spikes[2]. Gn is a key determinant of viral infectivity, and it is a major target for neutralizing antibodies, making it an attractive candidate for vaccine development and antiviral drug targeting[1][4]. Recombinant forms of Gn are used as diagnostic antigens to distinguish infected from vaccinated or uninfected animals (DIVA assays)[1]. No specific FDA-approved drugs target Gn, but it remains a leading antiviral research target due to its critical function in the RVFV life cycle[4].
