RIMS binding protein 3 (RIMBP3) is a scaffold protein and probable component of the manchette, a microtubule-based structure critical for sperm head morphogenesis during the later stages of sperm development[3][4][9]. RIMBP3 is part of the RIM-binding protein family, which includes proteins essential to presynaptic active zone organization and neurotransmitter release, but in mammals, RIMBP3’s main established function is in spermatogenesis rather than in synaptic transmission[2][3]. It is predicted to be involved in fertilization and spermatid development and localized to subcellular structures such as the manchette and the nucleus[3][5]. Mutations in RIMBP3 have been associated with male infertility, and genetic variants are linked to conditions such as Martsolf syndrome 1 and sulfonamide allergy[3]. RIMBP3 shares structural similarity with other RIMBP proteins, contains SH3 domains and fibronectin type III motifs, and has several paralogs including RIMBP3B and RIMBP3C[3][7]. There are no known drugs targeting RIMBP3, and it is not considered a therapeutic target at this time.
