Ring finger and CHY zinc finger domain containing 1 (RCHY1) is an E3 ubiquitin ligase that facilitates the ubiquitination and proteasomal degradation of target proteins including p53, HDAC1, CDKN1B, and TP73. It is a short-lived, nuclear-localized protein subject to self-ubiquitination and degradation, and it plays critical roles in the regulation of cell cycle progression, DNA damage response, and protein quality control. RCHY1 interacts with various transcription factors (notably HOX family proteins), and its dysregulation is implicated in cancer, where it can modulate the stability of key tumor suppressors. RCHY1 is being studied as a potential therapeutic target, particularly for cancers characterized by aberrant ubiquitin-mediated degradation of p53
Drugs theoretically targeting RCHY1 would act by inhibiting its E3 ligase activity; reducing ubiquitination and degradation of tumor suppressor proteins like p53, leading to increased tumor suppressor function and decreased proliferation
