Overview

Ring finger protein 121 (RNF121) is a transmembrane E3 ubiquitin ligase containing a RING finger domain, localized predominantly to the endoplasmic reticulum and Golgi apparatus in mammalian cells. RNF121 is evolutionarily conserved from C. elegans to humans and has been shown to regulate protein ubiquitination and degradation, specifically targeting immature membrane proteins such as vascular endothelial growth factor receptor-2 (VEGFR-2), integrin β subunits, and oncogenic proteins including MYCN. By controlling receptor maturation, trafficking, and signaling, RNF121 modulates angiogenesis, cell cycle, cell migration, and cellular response to stress. High RNF121 expression is associated with poor prognosis in MYCN-driven neuroblastoma, positioning it as a novel therapeutic target for cancer drug development and as a negative regulator of pathologic angiogenesis.

Mechanism of action

Drugs targeting RNF121 would likely inhibit its E3 ligase activity, leading to reduced stabilization of oncogenic proteins such as MYCN, increased cell surface VEGFR-2, and altered angiogenesis/cell adhesion. Interruption of ubiquitination may affect ERAD, protein quality control, and downstream pathological signaling.
