Overview

Ring finger protein 138 (RNF138) is an E3 ubiquitin-protein ligase that mediates protein ubiquitination, thereby signaling proteins for degradation via the proteasome. It is involved in DNA damage response and repair by promoting homologous recombination, regulates transcription by targeting key transcription factors like C/EBPα, and is implicated in the differentiation arrest of myeloid cells in acute myeloid leukemia. RNF138 also modulates the NF-κB signaling pathway, with downregulation linked to poorer prognosis and increased tumor progression in diseases such as colorectal cancer. RNF138 is a member of the RING finger protein family characterized by the RING domain, essential for substrate protein recognition and E2-E3 ligase activity. Its dysfunction is associated with cancer, immune dysregulation, and potentially neurodegenerative conditions.
