Overview

Ring finger protein 214 (RNF214) is a member of the RING finger ubiquitin ligase family, a group of enzymes that label substrate proteins with ubiquitin, thereby influencing their function, interactions, or localization[1][2][4][5]. RNF214 induces non-proteolytic ubiquitylation of TEAD transcription factors, thereby enhancing their interaction with YAP/TAZ and increasing the expression of Hippo pathway target genes[1]. RNF214 is involved in cell proliferation and is notably overexpressed in hepatocellular carcinoma, where it is associated with poor differentiation and reduced patient survival, acting as a potential oncogene and candidate therapeutic target in this cancer type. Its broader physiological and pathological roles remain understudied, but available evidence implicates RNF214 in general processes of transcriptional regulation, cancer biology, and possibly in other tumor types[1][6].
