Ring finger protein 215 (RNF215) is a human protein-coding gene located on chromosome 22q12.2 encoding a putative multispanning membrane protein of 377 amino acids, characterized by a RING finger zinc-binding domain. This domain suggests E3 ubiquitin ligase activity, implicated in protein ubiquitination and degradation, and possibly in trafficking proteins between the Golgi, endosome, and vacuole compartments[1][3]. RNF215 is overexpressed in several malignancies such as colorectal and head and neck cancers, where high expression predicts poorer overall survival and greater lymphatic invasion, making it a potential prognostic biomarker and therapeutic target. Mechanistically, RNF215 has been shown to regulate immune microenvironment composition and angiogenesis-related gene expression in tumor tissues. It is also a negative regulator of type I interferons in immune signaling. There are currently no drugs or inhibitors that specifically target RNF215, but its expression profile and functional properties suggest a potential role in cancer prognosis and therapy[1][3].
In principle, compounds could target RNF215’s E3 ligase activity or its pathway interactions, but as of now, no drugs are known to specifically target RNF215[1].
