Ring finger protein 223 (RNF223) is a member of the RING finger family of E3 ubiquitin-protein ligases, which catalyze the transfer of ubiquitin from E2 enzymes to specific substrate proteins, thereby regulating their degradation, stability, and cellular localization[4][5]. RNF223 is upregulated in pancreatic cancer, where it acts as an oncogene, promoting tumor growth and cell migration through effects on cancer cell metabolism and proliferation[1][2]. Its expression appears to be upregulated by the transcription factor KLF4. RNF223 is considered an independent prognostic marker in pancreatic cancer and may contribute to the pathogenesis of other malignancies, such as uterine sarcoma and endometrial cancer[2]. No approved drugs currently target RNF223 directly, and its clinical relevance is emerging in the context of cancer biology.
