Ring finger protein 39 (RNF39) is an E3 ubiquitin ligase encoded by the RNF39 gene located within the major histocompatibility complex class I region on chromosome 6. It contains a RING finger domain, typically involved in mediating protein-protein interactions and facilitating ubiquitin transfer. Evidence from rodent models and human data suggests it functions in early synaptic plasticity and modulates innate immune responses by ubiquitination of key antiviral adapters (e.g., DDX3X for Lys-48-linked and STING for Lys-63-linked ubiquitination). This dual role allows RNF39 to act as both an inhibitor and enhancer of antiviral signaling pathways. The gene is associated with human diseases such as psoriasis (by genetic association) and exhibits altered epigenetic status in multiple sclerosis. While highly expressed in brain tissue, particularly hippocampus, RNF39 is not currently recognized as a direct therapeutic drug target, nor are any small molecules known to modulate its function in clinical contexts[1][2][3][4][5][8].
