Ring finger protein 43 (RNF43) is a RING-type E3 ubiquitin ligase that acts as a negative regulator of the Wnt signaling pathway. It promotes the ubiquitination and endocytosis of frizzled receptors, leading to their removal from the cell surface and downregulation of Wnt signaling. Mutations in *RNF43* are frequently found in various cancers, and its altered expression may impact DNA repair capacity.
RNF43 mutations can sensitize cancer cells to Wnt antagonists by increasing Wnt pathway dependency.
