Overview

RING1 and YY1-binding protein (RYBP), also known as Death effector domain-associated factor (DEDAF), is a multifunctional protein that plays a critical role in both transcriptional regulation and the execution of apoptosis (UniProt Q8N488). As a member of the Polycomb group (PcG) proteins, RYBP is a core component of non-canonical Polycomb Repressive Complex 1 (PRC1), where it facilitates histone H2A ubiquitination and gene silencing (PubMed: 25483080). In the context of cell death, RYBP/DEDAF interacts with death effector domains (DEDs) of proteins like FADD and procaspase-8, significantly enhancing the assembly of the death-inducing signaling complex (DISC) and promoting apoptosis (PubMed: 11084014). Due to its frequent downregulation in various malignancies, including lung and liver cancers, it is primarily characterized as a tumor suppressor (PubMed: 25483080). While there are currently no FDA-approved drugs specifically targeting RYBP, its central role in epigenetic regulation and apoptotic pathways makes it a subject of interest for therapeutic strategies aimed at restoring its expression or activity in cancer cells (NCBI Gene ID: 23429).
