It functions primarily as a negative regulator of transcription, and is essential for proper cardiac development via its repression of the cardiac transcription factor TBX1. Mutations or loss-of-function of RIPPLY3 are associated with conotruncal heart defects, suggesting a critical role in cardiac morphogenesis and embryonic development. RIPPLY3 is predicted to localize to the nucleus, where it mediates transcriptional repression by RNA polymerase II.
