RNA, Ro60-associated Y1 (RNY1) is a small cytoplasmic noncoding RNA that forms part of the Ro ribonucleoprotein (Ro RNP) complex by tightly associating with the Ro60 protein and other partners[1][2][7]. RNY1 is evolutionarily conserved and is crucial for the structural assembly and function of Ro RNPs. It plays diverse roles such as regulating the subcellular localization of Ro60, scaffolding interactions with effector proteins, and influencing RNA quality control and degradation pathways[1][3]. RNY1 is not a traditional therapeutic target, but its complex (Ro RNP) is a major autoantigen recognized by autoantibodies in systemic lupus erythematosus and Sjögren syndrome, making the RNP complex clinically significant in autoimmunity[1][7]. Four human Y RNAs are known (Y1, Y3, Y4, and Y5), with Y1/RNY1 often functioning distinctly by recruiting or scaffolding various RNA-binding proteins[3][4]. Its structure features a conserved stem for Ro60 binding and additional modules that regulate protein partnerships and subcellular localization[1][3]. Direct pharmacological targeting of RNY1 itself is not currently described.
