The canonical U7 small nuclear RNA (snRNA) is an essential component of the U7 small nuclear ribonucleoprotein complex (U7 snRNP), primarily involved in the processing of histone pre-mRNA, specifically by facilitating the cleavage of the 3′ end of replication-dependent histone transcripts[1][5]. U7 snRNA recognizes a conserved downstream sequence (histone downstream element, HDE) to aid in precise histone mRNA maturation[1]. However, RNU7-153P is annotated as a pseudogene, meaning it is a DNA sequence similar to a functional gene but is considered non-functional due to mutations, truncations, or lack of necessary regulatory sequences[4][5]. There is no evidence that RNU7-153P produces a functional RNA product or participates in histone mRNA processing, nor is it regarded as a therapeutic target, disease gene, or biomarker[4][5]. Pseudogenes in the U7 family are common in primate genomes due to retrogene formation and gene duplication, but only the canonical U7 snRNA (for example, RNU7-1) is functional[5]. No disease association, interacting drugs, or safety considerations have been reported for RNU7-153P. The existence of RNU7-153P is primarily a consequence of gene duplication/evolutionary processes, and it should not be confused with the functional U7 snRNA required for histone RNA processing[1][5].
