Overview

RNA-binding motif protein 15B (RBM15B) is a member of the SPEN family of RNA-binding proteins, characterized by three RNA recognition motifs (RRMs) and a C-terminal SPOC (Spen orthologue and paralogue C-terminal) domain[1][2][5]. RBM15B participates in multiple aspects of post-transcriptional gene regulation, including alternative splicing, mRNA nuclear export, and guiding the m6A methyltransferase complex to specific mRNA sites for N6-methyladenosine (m6A) modification[1][2][4][5]. It operates as a cofactor for the nuclear export receptor NXF1 and interacts with proteins such as WTAP, METTL3, and METTL14, enabling site-specific methylation of mRNA[1][2][4]. RBM15B plays important roles in hematopoiesis (including B-lymphocyte and hematopoietic stem cell development), cellular differentiation, and has been implicated in the pathogenesis of various cancers, including leukemia, hepatocellular carcinoma, and lung cancer[1][2][5]. Abnormal expression or copy number variations of RBM15B are associated with altered patient outcomes in several malignancies[1][5]. As a non-enzymatic RNA-binding cofactor, RBM15B is not currently considered a direct therapeutic target, nor are any drugs known to directly interact with it[1][5]. It can serve as a component of biomarker panels for cancer prognosis based on m6A modification status and RBM15B expression[1].
