Overview

RNA binding motif protein 3 (RBM3) is a highly conserved, cold-inducible RNA-binding protein of the glycine-rich RNA-binding protein family. RBM3 contains RNA recognition motif (RRM) domains that enable it to regulate mRNA metabolism, including biogenesis, stability, splicing, export, and translation. It is strongly upregulated during physiological stresses such as hypothermia, hypoxia, and other adverse conditions, where it protects cells by promoting protein synthesis, encouraging cell proliferation, and preventing apoptosis. RBM3 has established roles in neuroprotection and is a key mediator of adaptive responses in the brain and muscle. In oncology, RBM3 acts as a proto-oncogene in several tumor types and is under investigation as a diagnostic and prognostic biomarker. Experimental data demonstrate that RBM3 is involved in regulating cell polarity, morphology, and migration via the RhoA-ROCK-CRMP2 signaling pathway. Despite its importance as a cellular protective factor and cancer biomarker, it is not currently a direct drug target; rather, RBM3 expression is monitored for disease association or as a potential therapeutic marker.
