RNA-binding motif protein 33 (RBM33) is a nuclear RNA-binding and RNA-reader protein that facilitates the export of a broad spectrum of GC-rich RNA transcripts from the nucleus into the cytoplasm, by directly binding to these RNAs and recruiting them to the TREX–NXF1/NXT1 RNA export machinery. It contains a carboxyl-terminal RNA recognition motif (RRM) that mediates specific m6A binding and regulates the activity and substrate selectivity of the RNA demethylase ALKBH5, thereby impacting mRNA stability and autophagy. RBM33 is highly expressed in various cancers, particularly head and neck squamous cell carcinoma, where it drives tumorigenesis by stabilizing DDIT4 mRNA in an m6A-dependent manner and promoting autophagy. It predominantly localizes to the nucleus, shuttling between the nucleus and cytoplasm, and is active in both compartments. RBM33 is not a known drug target, but its interaction with the ALKBH5 demethylase and the TREX export pathway reveals important regulatory functions in RNA processing relevant to gene expression and cancer biology.
